I use the MyIDE+Flash IDE Interface (Cartridge) to flash OSb into my 800xl. The 800xl will boot OSb without the interface cartridge plugged in or does the cartridge needs to remain plugged in to run new OS?

The MyIDE+Flash cartridge has it's own OS that loads into the 800xl. Any other OS will be in conflict to drive the IDE-port. So when you boot the system, first the MyIDE OS will be loaded and continues the boot-process. If an user then directs to load OSb, the MyIDE-OS will be overwritten.
You can how ever load a OSb version with patched IDE-drivers, for this the cartridge memory must be cleared. Or you can re-programm the cartridge to do this for you (see atarimax.com for detailed intructions).
The MyIDE cartridge is not a 'pass-through' device and so you wouldn't be able to play any other cartridge at the same time. If you have a particular title in mind then searching here or on AtariAge would most likely find a 'fixed' version in executable form (i.e. so that it could be loaded from disk/flashcart) that can be installed on a hard disk/compactflash and run using your MyIDE+Flash cartridge. The fixes also take care of the XL compatibility in all but a few cases.

With the internal MyIDE-version (needs soldering) you keep the cartridgeslot free. Some 400/800 carts work with this OS, I have put some fixes in. Demon-attack is one for sure, I like this game and patches the OS for it
